KABUKSUZ ÇEKİRDEK KABAKLARINDA (Cucurbita pepo L. var. styriaca) IŞINLANMIŞ POLENLE TOZLAMA YÖNTEMİ KULLANILARAK HAPLOİD ÜRETİMİ Production of haploids in naked seed pumkins by pollination with irradiated pollen Meltem BERBER Kazım ABAK ABSTRACTThis study is realiesed for two basic purposes. First one is to produce haploid plants from naked seed pumpkins (Cucurbita pepo var. styriaca) with irradiated pollination technique and the second one is to research the most suitable doses of irradiation for this aim. In the research the shelled seed pumpkin genotypes are also used as control and a total of fifteen genotypes were used as plant matarial. Three irradiation dosses of gamma rays (50, 100 and 150 Gray) were tested. During the study, a total of 2073 embryos are rescued from different genotypes and 979 of these embryos transformed into plants. Haploid embryos are obtained in all of the used material and any important difference has not been observed between genotypes. All three dosses of irradiation gave good results in the study. However; more haploid plants are acquired from the 150 Gray. 75 of plants were acclimatized and cultivated in a plantichimes and ploidy level of there plants were determined using indirect methods (existence of pollens, leaf and flower features, stoma density and chloroplast number in gard cells of stoma). 42.6% of these plants seen to be haploid and 57.3% of them diploid.
